Introduction to vitrification cryogenic liquid

Vitrified cryogenic liquid, also known as vitrified coolant, is a liquid used to freeze and preserve biological samples. Its main feature is its ability to form a solid structure similar to a glass state at extremely low temperatures, thus preventing the movement of active molecules within biological samples and allowing the samples to remain intact for a long time.

 

Vitrification fluid usually consists of one or more organic solvents and one or more protective agents. Commonly used organic solvents are dimethyl sulfoxide (DMSO) or ethylene glycol, while protective agents include sucrose, glycerin, etc. These ingredients form hydrogen bonds with water molecules and prevent them from crystallizing, thus protecting the integrity of cells or tissues.

 

The mechanism of action of vitrification liquid is to directly transform the sample from liquid state to glass state by lowering the temperature at which ice crystals form. Before the sample is frozen, the vitrification liquid will gradually reduce the sample to an extremely low temperature (usually around minus 100 degrees Celsius) through a series of cooling processes. During this process, active molecules within the sample are immobilized, reducing chemical reactions and biodegradation.

 

Vitrification fluid is widely used in the long-term preservation and transportation of biological samples such as cells, tissues, and blood. It can effectively maintain the integrity and stability of the sample, allowing the sample to return to its pre-frozen state after thawing. In addition, vitrification liquid can also be used in applications such as ultra-high-resolution imaging, protein crystallization, and dry preservation of cryopreserved biological samples.
