1. Hybridization temperature selection: The selection of hybridization temperature depends on the length of the probe, the complexity of the sequence, the number of interference bands in the sample, the strength of the background signal in the bottom band, and other factors. Generally, when hybridizing shorter single-stranded probes, a higher temperature can complete the hybridization reaction.

2. Post-hybridization processing: Washing the membrane is a necessary post-processing step to obtain positive hybridization bands. Usually the buffer used when washing the membrane is 6×SSC and the temperature is 50°C.

3. Hybridization reaction time: Hybridization reaction time varies depending on factors such as temperature, hybridization probe length, specificity, and background signal strength in nucleic acid samples. The generally recommended reaction time is 16-24 hours.
