Micromanipulation fluid
Micromanipulation fluid is used for micromanipulation, which can provide protection for organisms in a hypotonic environment and prevent cells from absorbing water and bursting. It is mainly used for micromanipulation of mammalian embryos, such as in vitro maturation culture of oocytes, fertilization operations, embryo operations, etc.
The composition of the micromanipulation fluid includes the following ingredients:
1. Basal fluid: TCM-199 (a complete cell culture medium for mice) is commonly used, containing mouse serum, amino acids, glucose, inorganic salts, etc.
2. Osmotic pressure regulator: 4.5% NaCl solution is commonly used.
3. Cell metabolism inhibitors: such as polysorbate 80 (a non-ionic surfactant) and heparin sodium (an anticoagulant), can inhibit cell metabolism and prolong cell life.
4. Cytostatics: such as mitomycin C and cyclophosphamide, which can inhibit cell growth.
5. Other additives: such as antibiotics, vitamins, etc.
The use of micromanipulation fluid includes oocyte maturation culture, fertilization operation, embryo operation, etc. It should be filtered and packaged before use to remove bubbles and impurities. During use, the temperature and pH value should be kept stable and the fluid should be replaced regularly.
